Russia’s first serially produced open DNA sequencer
NANOPHOR 05
An eight-capillary genetic analyzer for Sanger sequencing and DNA fragment analysis with laser-induced fluorescence detection.
- 8 capillaries
- 7 detection channels
- Open system
Developed by SYNTOL together with the Institute for Analytical Instrumentation of the Russian Academy of Sciences. Serially manufactured in Russia since 2015.
Key specifications
Eight capillaries, seven channels and a 96-well plate
parallel sample separation
0.2 ml plate format
fluorescent-dye detection
read length at no less than 98.5% accuracy
Two analytical modes
Sanger sequencing and DNA fragment analysis
The analyzer separates fluorescently labelled DNA fragments in a polymer matrix and records the signal in seven detection channels. Results can be exported as .ab1 and .fsa files.
Sanger sequencing
DNA Analysis software converts sequencing-reaction peaks into a nucleotide sequence and produces an .ab1 file.
Fragment analysis
DNA FA software determines fragment lengths and alleles using size standards, bin panels and allelic ladders, then produces an .fsa file.
Instrument and software
Instrument and software for data acquisition and analysis
Eight capillaries
The analyzer uses a 96-well plate and separates samples in eight capillaries in parallel.
Seven detection channels
The optical system records fluorescent dyes in seven spectral channels.
Four software applications
The software set includes instrument control, DNA Analysis, DNA FA and the PAR²SEQ sequence editor.
Standard data formats
Export to .ab1 and .fsa allows further processing in software that supports these formats.
Operating principle
How the analytical result is produced
-
01
Sample loading
Prepared reactions are placed in a 96-well plate.
-
02
Electrokinetic injection
DNA fragments are introduced from the wells into the capillaries by an electric field.
-
03
Separation
Fragments are separated by length in a polymer matrix under high voltage.
-
04
Fluorescence detection
A 488 nm laser excites the dyes and the optical system records the signal.
-
05
Data processing
Software produces a DNA sequence or a fragment profile.
Example data
Results obtained using NANOPHOR 05
STR fragment analysis
A VeriFiler™ Express positive control processed in software for STR-profile analysis.
ITS-region sequencing
The Glycyrrhiza inflata ITS region was sequenced using PDMA-6 polymer and 50 cm capillaries.
