Biotinylated oligonucleotides
Biotin has high affinity for avidin and streptavidin, so biotinylated oligonucleotides are used in hybridization, blotting, sequencing, genotyping and immobilization applications.

Applications
Biotin has high affinity for avidin and streptavidin. Biotinylated oligonucleotides are therefore used for:
- in-situ hybridization;
- Southern blotting;
- chemiluminescent sequencing;
- analysis of point mutations;
- protection of the oligonucleotide 3′ end;
- genotyping;
- cloning of unknown DNA sequences;
- immobilization on solid supports.
Purification and yield
Biotin is introduced at either the 5′ or 3′ end directly during synthesis. Biotinylated oligonucleotides are purified by reverse-phase HPLC or polyacrylamide gel electrophoresis.
- Guaranteed minimum yield after purification at a 250 nmol synthesis scale: 4 optical density units (20 nmol).
- Guaranteed oligonucleotide purity: at least 95%.
