Optical-density units

OD describes measured material but is not a fixed molecule count

Oligonucleotides absorb UV light at 260 nm. Conversion from absorbance to molar and mass concentration depends on composition, sequence, molecular weight, and modifications.

A260 primary measurement

Absorbance is measured at 260 nm using a known optical path.

ε extinction coefficient

It is calculated for the specific sequence and base order.

µM molar concentration

Represents the number of molecules per solution volume.

µg/µL mass concentration

Uses the molecular weight of the complete construct.

Understanding OD

Optical density is linked to solution absorbance

For a 1 mL solution and 1 cm optical path, A260 is used as a quantitative oligonucleotide measure.

Sequence matters

Purines, pyrimidines, and nearest-neighbor stacking change absorbance.

Modifications matter

Labels may contribute to molecular weight and optical properties.

OD is not directly nmol

Equal absorbance can represent different substance amounts for different sequences.
