Summary

Linear probes may carry the fluorophore and quencher at opposite ends or include one of the labels inside the oligonucleotide chain. The text explains the main probe configurations, the effect of terminal guanosine and quencher position, and cases where internal labeling is preferred, for example for longer probes or lower background fluorescence.
